JewelStaiteFan, you beat me to it. My favourite line from Aliens:

Burke: This facility represents a significant monetary value to the company.

Ripley: They can bill me!

Everything you need to know about Ellen Ripley in four words.

But, coming in a close second:

Ripley: Show me everything. I can handle myself.

Hicks: Yeah, I noticed.

It's such a great line in such a great movie!

Funny though, how Doc Ostrow was able to "figure out" the whole Krell Machine mystery with just one jolt to the brainpan, whereas Morbius couldn't figure it out for 18 years.

Well, it's not a single line, but I submit this from 17 Again, just 'cause Zac Efron was Young Simon...

Ned Freedman: It's a classic transformation story. Are you now or have you ever been a Norse God, Vampire, or Time-Traveling Cyborg?
Mike O' Donnell: I have known you since what, first grade? I think that maybe I would have told you!
Ned Freedman: Vampire wouldn't tell. Cyborg wouldn't know.

From the script for...

Plan 9 from Outer Space

[TITLE: "Criswell Predicts..."]

CRISWELL
Greetings, my friend. We are all interested in the future, for that is where you
and I are going to spend the rest of our lives. And remember my friend, future
events such as these will affect you in the future.
You are interested in the
unknown, the mysterious, the unexplainable. That is why you are here. And now,
for the first time, we are bringing to you the full story of what happened on
that fateful day. We are giving you all the evidence, based only on the secret
testimonies of the miserable souls who survived this terrifying ordeal. The
incidents, the places, my friend we cannot keep this a secret any longer. Let us
punish the guilty, let us reward the innocent. My friend, can your heart stand
the shocking facts about grave robbers from outer space?

[TITLE SEQUENCE]

[SMALL FUNERAL, EVERYONE COMPLETELY SILENT]

CRISWELL
All of us on this earth, know that there is a time to live, and that there is a
time to die, yet death is always a shock to those left behind. It is even more
of a shock, when death, the proud brother, comes suddenly without warning. Just
at sundown, a small group, gathered in silent prayer around the newly-opened
grave of the beloved wife of an elderly man. Sundown of the day, yet also the
sundown of the old man's heart, for the shadows of grief clouded his very
reason. The funeral over, the saddened group left the graveside. It was when the
gravediggers started their task that strange things began to take place.

*************
There's a reason why Plan 9 is called the worst movie ever made. The dialogue is a big part of it.

"Might have been the losing side, still not convinced it was the wrong one." -Mal

... fully loaded, safety off...

Some great lines... fwiw I have added a bunch to the scifiradio.com playlist - love the Pitch Black ones especially - that's such an underrated movie. Vin kind of cheeses it a bit and the ending feels a little forced... and the creature effects are showing some age... but it's a fun scifi flick. Cole Hauser as Johns and Radha Mitchell as Fry are great.

Also - in the course of tracking down lines I came across this site:

http://en.wikiquote.org/wiki/Main_Page

You can spend a lot of time there! Caution: If you are a stickler for getting quotes exact know that I have found some of them to be inaccurate.

Okay, well, if we're supposed to be going for a great single line, Stargate SG-1 was a treasure trove...

You think you know--what's to come, what you are. You haven't even begun.

Well, it isn't sci-fi per se, but since I think it's one of the most quotable movies ever, I must submit some Princess Bride:

"You keep using that word. I do not think it means what you think it means."

"Rodents of unusual size? I don't think they exist."

"There's a shortage of perfect breasts in the world. 'Twould be a pity to damage yours."

"We are men of action. Lies do not become us."

"Can you move at all?"
"Move? You're alive. If you want I could fly."

"You're trying to kidnap what I've rightfully stolen."

"Your ears you keep, and I'll tell you why. So that every shriek of every child at seeing your hideousness will be yours to cherish. Every babe that weeps at your approach, every woman who cries out, "Dear God, what is that thing?" will echo in your perfect ears."

"I admit it, you are better than I am."
"Then why are you smiling?"
"Because I know something you don't know."
"And, what is that?"
"I am not left handed."

"Have fun stormin the castle!"

"I can cope with torture."

"Truly, you have a dizzying intellect."

"You seem a decent fellow. I hate to kill you."
"You seem a decent fellow. I hate to die."

"I'm not a witch, I'm your wife!"

"I do not envy you the headache you'll have when you awake. Until then rest well, and dream of large women."

"I didn't mean to jog him so hard."

"You'll put down your rock, and I'll put down my sword, and we'll try to kill each other like civilized people?"

"Fezzik, are there rocks ahead?"
"If there are, we all be dead."

"Drop. Your. Sword."

I could go on for days, so I should really just stop. And yes, I know I left a huge favorite out. I left it out because it's such a ridiculously huge favorite.

[trying to explain to Captain Bipto that their invasion has failed]
Giggywig: Let me sum up the entire situation in a nutshell: there are five of us, and four billion of them. They have Strategic Air Command, nuclear powered submarines, and John Wayne. We have this.
[holds up a small rifle]
Captain Bipto: Is it loaded?
[points the rifle at Bipto]
Giggywig: Let's find out!

Right on, Spaced Invaders, one of the worst, funniest sci-fi movies ever made. a personal fave, guilty pleasure movie. Gotta dig it out , it's a Halloween movie. I was gonna post " Prepare to die, Earth scum!" myself.

How 'bout: "We're toast. In your case , a whole loaf of toast."

Alright children, look both ways before crossing the street.
If only captain Bipto had known!

I pray for one last landing,
on the globe that gave me birth.
Let me rest my eyes on the fleecy skies
and the cool, green hills of Earth.
