Man, it's hard to choose; there's so many (even limiting myself to the actual fighters [as opposed to the strategists, commanders, leaders, and iron-willed adventurers]). Some of my more obscure favourites include:


Miriya Parino/Sterling, ace pilot, field leader of the Zentraedi's Quadrono Battalion, defector, and second-in-command of Earth's Skull Squadron, wife of Max, mother of Dana & Aurora, and probably the first real "amazon" I ever encountered. This is her strapping into her Quaedlun-Rau (basically a weaponized space suit) in Robotech/Macross.


Klan Klan. Miriya's quasi-heir, from Macross Frontier. Also a Quaedlun-Rau pilot, commander of SMS' Pixie Squadron (heirs to the Quadrono tradition). For those who've never met the Zentraedi, that tiny little thing she's looking at is a human in the foreground. T'sentrati are BIG.


Deunan Knute. As envisioned by Shinji Aramaki in Appleseed, she is bar-none the best fighter I have ever seen anywhere --a natural acrobat, sharpshooter, ninja, stunt-driver, and coordinated enough to use the recoil from her weapons to shift positions in mid-air. She could kick Vin Diesel's ass.


Major Motoko Kusanagi, from Ghost in the Shell. The most skilled proshetic body operator and web-diver alive. A "cyborg" with no organic parts --everything on/in her came from a factory except her consciousness. Her body is just another suit of clothes to wear.


Lt. Cmdr. Domenica "Core-Breach" Corsi of the USS daVinci, from Star Trek: SCE/Corps of Engineers. She's basically the Star Trek equivalent of Zoe (and, like Zoe, she's romantically linked to a loveable goofball genius).

Well, River, of course (she did it without guns, remember, 99% of the time!), and personally I'd say Ripley #2.

Buffy? She was a woos...yeah, so she killed vampires, but she had SUPER STRENGTH to do it and still got her ass kicked a lot. Besides, she spent seven years doing nothing but moaning about not having a life; that's a strong woman? Pffft.

Faith comes closer, but I don't like her politics or attitude.

River is a "pure" heroine...how many of those are running around? You ARE speaking of best "ass kicker", not strategist, thinker, plotter, etc., so it has to be River.
